    Mifepristone increases mRNA translation rate, triggers the unfolded protein response, increases autophagic flux, and kills ovarian cancer cells in combination with proteasome or lysosome inhibitors

    The synthetic steroid mifepristone blocks the growth of ovarian cancer cells, yet the mechanism driving such effect is not entirely understood. Unbiased genomic and proteomic screenings using ovarian cancer cell lines of different genetic backgrounds and sensitivities to platinum led to the identification of two key genes upregulated by mifepristone and involved in the unfolded protein response (UPR): the master chaperone of the endoplasmic reticulum (ER), glucose regulated protein (GRP) of 78 kDa, and the CCAAT/enhancer binding protein homologous transcription factor (CHOP). GRP78 and CHOP were upregulated by mifepristone in ovarian cancer cells regardless of p53 status and platinum sensitivity. Further studies revealed that the three UPR-associated pathways, PERK, IRE1α, and ATF6, were activated by mifepristone. Also, the synthetic steroid acutely increased mRNA translation rate, which, if prevented, abrogated the splicing of XBP1 mRNA, a non-translatable readout of IRE1α activation. Moreover, mifepristone increased LC3-II levels due to increased autophagic flux. When the autophagic–lysosomal pathway was inhibited with chloroquine, mifepristone was lethal to the cells. Lastly, doses of proteasome inhibitors that are inadequate to block the activity of the proteasomes, caused cell death when combined with mifepristone; this phenotype was accompanied by accumulation of poly-ubiquitinated proteins denoting proteasome inhibition. The stimulation by mifepristone of ER stress and autophagic flux offers a therapeutic opportunity for utilizing this compound to sensitize ovarian cancer cells to proteasome or lysosome inhibitors.Fil: Zhang, Lei.     Results of Large Area Crop Inventory Experiment (LACIE) drought analysis (South Dakota drought 1976)

    LACIE using techniques developed from the southern Great Plains drought analysis indicated the potential for drought damage in South Dakota. This potential was monitored and as it became apparent that a drought was developing, LACIE implemented some of the procedures used in the southern Great Plains drought. The technical approach used in South Dakota involved the normal use of LACIE sample segments (5 x 6 nm) every 18 days. Full frame color transparencies (100 x 100 nm) were used on 9 day intervals to identify the drought area and to track overtime. The green index number (GIN) developed using the Kauth transformation was computed for all South Dakota segments and selected North Dakota segments. A scheme for classifying segments as drought affected or not affected was devised and tested on all available 1976 South Dakota data.     The diplopod orders Callipodida and Polydesmida, and their respective families Abacionidae and Xystodesmidae, are initially recorded from South Dakota as is Polydesmidae from North Dakota. Other new records of indigenous taxa include Abacion Rafinesque, 1820/A. texense (Loomis, 1937) and Pleuroloma/P. flavipes, both by Rafinesque, 1820, from South Dakota, and Pseudopolydesmus Attems, 1898/P. serratus (Say, 1821) from Alabama, Connecticut, Delaware, New Hampshire, North Dakota, South Carolina, and the District of Columbia. New records of Aniulus garius Chamberlin, 1912, A. (Hakiulus) d. diversifrons (Wood, 1867), and Oriulus venustus (Wood, 1864) (Julida: Parajulidae) are provided for western Minnesota and/or eastern North Dakota. Published records from these states are summarized, and the introduced taxa, Julidae/Cylindroiulus Verhoeff, 1894/C. caeruleocinctus (Wood, 1864) and Paradoxosomatidae/Oxidus Cook, 1911/O. gracilis (C. L. Koch, 1847), are newly recorded from the Dakotas. The distribution of P. serratus, which extends from Maine to South Carolina and the Florida panhandle, west to Texas, and north to Fargo, North Dakota is described and discussed. This distribution exhibits a prominent southeastern lacuna which we hypothesize suggests replacement by younger, more successful species, as postulated for a similar distributional gap in Scytonotus granulatus (Say, 1821)

    Nearly 40% of South Dakota agricultural land is operated under a leasing agreement. Presented in this report are recent and longer term trends in land tenure, ownership, and leasing, based on Census of Agriculture data and related materials. Also presented are findings from the 1996 farmland leasing survey completed by 513 South Dakota farm operators: (1) characteristics of rental market participants and of the farmland leasing market, (2) detailed provisions of cash leases and share leases for cropland, and (3) economic evaluation of farmland leasing arrangements. Information from the 1996 survey provides a comprehensive and statistically valid benchmark of agricultural land leasing in South Dakota, with primary emphasis on cropland leasing arrangements. This is the most comprehensive statewide study of South Dakota farmland rental markets since 1986. In many respects this publication updates and extends findings from the 1986 study reported in B 704, Farmland leasing in South Dakota, published by the South Dakota Agricultural Experiment Station (Peterson and Janssen, 1988).     Higher Education on Buildings: Case Study in the North Dakota Region

    Because of the growing demand for local skilled professionals to improve the health, energy efficiency, and sustainability of residential and commercial buildings in North Dakota, this case study reports the current situation of higher education relating to buildings in the state’s vicinity, including Minnesota, Montana, North Dakota, and South Dakota. In this region, 116 programs relating to buildings were found in 41 postsecondary institutions, and both their majors and courses were then studied with frequency lists. The frequency information was analyzed over nine sets of curriculum areas at both graduate and undergraduate levels for the four states. After the current state of buildings in North Dakota was investigated, strategies were then proposed to rectify current issues regarding higher education on buildings, including but not limited to forming a comprehensive and interdisciplinary program on buildings (e.g., architectural engineering), providing more graduate programs, developing more courses in areas that lack adequate coursework, and increasing student enrollment. These strategies will greatly promote the health, energy efficiency, and sustainability for new and existing buildings in the four-state region of Minnesota, Montana, North Dakota, and South Dakota
